To estimate the bystander effect in 3D cell systems, we have introduced Fucci (Fluorescent Ubiquitination-based Cell Cycle Indicator) cells for single cell targeting and tracking after irradiation. Fucci cells show specific colors of cell nuclei depending on cell cycle. We tried to develop a HeLa-Fucci spheroid as a 3D system to simulate in vivo tumor. In this study, we exposed the center of HeLa-Fucci spheroids using synchrotron X-ray beam and investigated the radiation effect on the cell cycles by a fluorescent live-cell imaging technique. The cell cycle arrest of non-irradiated "bystander" cells surrounding the exposed area was not significant. This might be attributed to faulty gap junctions between the cancer cells. Cell cycles of bystander cells could be affected when the HeLa-Fucci cells were mono-layered cultured, indicating that only medium transfer pathway of bystander cell cycle effect would be workable.
