Presentation Effects of carbon ion on expression of organic anion transporting polypeptides

The organic anion transporting polypeptides (OATPs) belong to the solute carrier family and is a group of membrane transporters that have been extensively recognized as key determinants of absorption, distribution, metabolism and excretion (ADME) of various drugs, xenobiotics and toxins. Many anti-tumor drugs are substrates of OATPs and Accumulating evidences in recent years have shown that OATPs distribute differently in tumor cells and/or tissues. Therefore, OATPs, especially the ones that were demonstrated to be able to transport anti-tumor drugs, have been proposed to serve as novel therapeutics targets for future drug discovery. Our previous study revealed that X-ray irradiation can increase expression level of OATP family member OATP1A2, an OATP that was found to transport antiturmor drugs, in breast cancer cells. The combined treatment with X-ray and OATP1A2 substrate methotrexate exerted a more significant effect on growth rate and cell death compared with irradiation or MTX treatment alone. In the current study performed at NIRS, we investigate the effect of high LET irradiation on expression of OATPs and two major efflux transporters ABCB1 and ABCG2.
