The first and the second order variations of the elastic free energy for a vesicle are obtained to study the stability of the equilibrium shape of the vesicle. We consider the membrane of the vesicle contain a certain amount of inclusions whose distribution changes along the membrane, that results in the non-uniform changes of the spontaneous curvature, the bending rigidity and the surface energy of the membrane. With the assumption that the vesicle has an axisymmetric shape, the explicit formulae of the variations are obtained. Although the formulae obtained are quite complicated ones, they are used to analyze the stabilities for two simple shapes, cylinder and sphere, as examples.
