INTEGRATION/ EXCISION MECHANISM OF SPβ PROPHAGE IN BACILLUS SUBTILIS

In Bacillus subtilis strain 168, the SPβ prophage is inserted into a spore polysaccharide synthesis gene spsM.The SPβ excision supports two pathways both the phage life cycle and normal sporulation to reconstitute the spsM gene of the host cells, and it requires a site-specific recombinase, SprA, and an accessory protein, SprB. In this study, I showed that the process of integration is catalyzed by SprA (integrase), whereas the excision is catalyzed by SprA but also requires SprB (RDF; recombination directionality factor). Furthermore, I demonstrated that the extracellular SPβ DNA released from the mother cell during sporulation can be horizontally acquired by recipients without SPβ prophage through the process of natural competence.
