Nicotine has been suggested that may directly act onCa2+ pump in sarcoplasmic reticulum and K+ channel of cardiomyocytes. To verify them, multi electrode array (MEA)system that can measure extracellular field potential was adopted. I explored the optimal medium conditions because MEA system protocols have not been established. As a result, extracellular field potential of cardiomyocytes was stably measured in DMEM with 25 mM HEPES. Therefore I used this medium for the examination of nicotine effect to cardiomyocytes. Nicotine shortened interspike interval and prolonged field potential duration. From these results, it was suggested that nicotine directly promoted the function of Ca2+ pump in sarcoplasmic reticulum and inhibited the function of K+ channel of cardiomyocytes.
